Wild introgression lines of Oryza nivara as potential donor for yield and seedling vigour traits and mapping associated genomic regions

Wild species and introgression lines are known for their survival under adverse conditions and have been explored for their unique genes that can broaden the genetic diversity as well as climate resilience. In our study, an advanced intercross population derived from wild introgression lines of Oryza nivara was explored to uncover novel QTLs for yield and seedling vigour related traits. Significant variation was observed in Oryza nivara derived advanced inter-cross population with high heritability and genetic advance for the traits under study. Further, we employed composite interval mapping to detect major QTLs linked to various yield and seedling vigour related traits. Among the 19 QTLs detected with PVE > 5%, two major QTLs were identified in chromosome 7. QTL for test weight qTW_7.1 was detected in the region of A07P06137-A07PP09975 with a PVE of 11.07% and QTL for panicle weight qPW7.1 was detected between RM18-RM478 with a PVE of 10.16%. Single marker analysis detected QTL i.e., qPH8.1 and qFRW11.1 for plant height and fresh root weight respectively, with PVE > 10%. As the major QTL (qTW7.1) between the marker interval of A07P06137-A07P09975 co segregates for grain yield and seedling vigour traits, the region was further fine mapped from 3.84 Mb to 199 kbp by genotyping by sequencing. Co-localization of several candidate genes associated with stress tolerance and grain yield has been reported in the fine-mapped chromosomal region, including OsFBX232, which encodes an F-box protein with SUMO/Centrin/UBL1 domains involved in protein regulation and stress response. The identification of genotypes exhibiting a combination of early seedling vigor, high yield potential, and QTL clusters for these traits underscores the utility of wild introgression lines in breeding programs.
