<p>Two new compounds (<b>1</b> and <b>2</b>) and six known compounds (<b>3–8</b>) were isolated from the endophytic fungus <i>Aspergillus fumigatus</i> J2-22-1, and their structures were elucidated through comprehensive NMR spectroscopy and HR-ESI-MS analyses. All compounds were evaluated for PTP1B inhibitory activity. Notably, compound <b>1</b> demonstrated significant inhibition with an IC<sub>50</sub> value of 7.36 μM, demonstrating comparable potency with the positive control Na<sub>3</sub>VO<sub>4</sub>. These findings underscore the potential of fungal metabolites as promising sources for developing novel PTP1B inhibitors.</p>
