Analysis of the Ciliotoxic Effects of an Intranasal In Situ Ribavirin Delivery System: In Vitro and In Vivo Evaluation

The intranasal route of drug administration offers a number of advantages, including the possibility of direct brain targeting while bypassing the blood–brain barrier. The local effect of an innovative ribavirin in situ gel on the nasal mucosa of outbred rats was investigated. Twelve formulations containing ion-sensitive polymers were evaluated in vitro and their physicochemical properties were assessed. The formulation containing 0.5% gellan gum and 2% Poloxamer-124 was recognized as optimal, exhibiting the most suitable technological characteristics. In vivo experiments confirmed the absence of irritating effect on the surface of the nasal mucosa in experimental animals. These findings highlight the potential of this dosage form for the treatment of brain tumors.
