Farmed Atlantic salmon (Salmo salar) skin mucosa during seawater transfer compared with wild juveniles

Skin mucous cell (MC) parameters were evaluated across five commercial trials with farmed Atlantic salmon (Salmo salar) during the seawater transfer (SWT) phase. In these anonymized dietary studies (n = 512), both MC area (MCA) and abundance declined following SWT and began to recover approximately 33–68 days post-transfer. The decline in MCA ranged from 16.5 to 51.9%, while the reduction in MC abundance ranged from 35.0 to 57.8% of initial values. This drop in skin MC parameters indicates a period of mucosal vulnerability followed by restitution. To assess whether this trend is reflected more broadly, we plotted the mean skin MC parameters from all 157 sampling events (defined as the sampling of salmon from a single production facility and rearing system on a given day; total n = 3430 skin samples, Quantidoc database), which revealed a generally U-shaped curvilinear relationship between fish weight and MC parameters in the salmon life cycle. A significant U-shaped quadratic relationship between MCA and the square of fish weight was found (lm, p < 0.05), with the lowest MCA values occurring between 150 and 250 g. This non-linear pattern suggests that skin MC dynamics in the salmon around SWT may reflect stage-specific physiological adjustments. In the skin, wild smolts tended to exhibit lower MC values than wild salmon adults, suggesting that the observed reduction and subsequent recovery in MC parameters could represent a conserved physiological response to seawater adaptation. Interestingly, wild smolts appeared to show lower values than farmed smolts, which may indicate possible adaptive differences between the populations.
