<p>Biofloc culture system has a several nutritional advantages as compared to traditional system in terms of essential nutrients with a single cell protein as well as assimilation of inorganic nitrogen by heterotrophic bacteria. Whereas, semifloc contains a periphytic substrate are consist of the algae, heterotrophic and autotrophic microbes, zooplankton and other invertebrates naturally available source of nourishment for the culture animal. Which will help to reduced dependency on the commercial feed and economical feasible compared to tradition farming. In present study, one twenty days of trial was conducted in HDPE polythene lined ponds with the treatments of semi-floc (SF = vertical periphytic substrate and lower carbon addition) and biofloc (BF = C:N ratio with periphytic substrate) and conventional system (Control = without biofloc and periphyton). The experiment was conducted at commercial farms located at Nagapattinam district, Tamil Nadu, India. <i>Penaeus vannamei</i> post larvae 58, 72 and 116 post larvaem<sup>2</sup> (Initial ABW = 0.086 ± 0.001&#xa0;g) were stocked in control, SF and BF ponds respectively. The microbial dominance revealed that heterotrophic bacterial community was significantly higher whereas vibrio load was decreased in the BF culture medium followed by SF groups than control. Cumulative mortality rate was lower in the BF, SF groups respectively. Total hemocyte count (THC), DHC (hyaline, semi /granular hemocytes) and phagocytotic hemocytes was increased in BF shrimps followed by SF shrimps than control (<i>P</i> &lt; 0.05). Phenoloxidase enzyme activity was higher in BF shrimp serum samples followed by SF treatments whereas positive control was recorded low. Consistently the lysozyme assay was showed decreased OD values in the BF groups when compared other treatments. The percentage inhibition of SOD level was higher in BF treatment groups (41.21—61.75%) followed by SF group whereas positive control (39.23 ± 1.99) was recorded lower. The cellular and humoral immune responses were enhanced in BF groups as compared to other treatments when challenged with pathogen <i>V. parahaemolyticus</i>. Hence, present study elucidated that SF and BF with periphyton based shrimp farming was significantly better one farming practices in terms of productivity, water quality, protective immune responses, economically viable and sustainable technology.</p>
