Influence of in vitro pulse treatments with plant growth regulators and seaweed derived bio-stimulant on the critical growth parameters of Gracilaria dura—a commercial agarophyte

The use of bio-stimulants is increasingly recognized as key to sustainable seaweed cultivation. This study explores the use of pulse treatments of various bio-stimulants and their combinations, designed to minimize overstimulation. This approach improves cost-efficiency and is well-suited for scaling up production of commercially important seaweeds. The present study investigates the effects of pulse treatments with bio-stimulants; 6-Benzylaminopurine (BAP), Kinetin (KIN), Gibberellic acid (GA3), and Ascophyllum Marine Plant Extract Powder (AMPEP) on survival, regeneration, accessory pigments, and antioxidant capacity of Gracilaria dura under in-vitro conditions. Pulse treatments significantly improved survival rates, especially the combination treatments, achieved near 100% survival, while regeneration rates were highest under 1-pulse conditions (KIN, GA3, and AMPEP; nearly 61%). Regeneration of lateral shoot per explant were observed higher with 3-pulse treatment of GA3 (1.51 ± 0.21%) followed by 1-pulse of KIN (1.47 ± 0.26%) and combination (1.46 ± 0.19%) treatments, respectively. Accessory pigments show variability across treatments. 2-pulse of KIN (59.25 ± 12.19 µg g−1 FW) and 3-pulse of GA3 (58.73 ± 12.20 µg g−1 FW) significantly increase the chlorophyll-a content, while the 6-pulse treatment of GA3 and 4-pulse treatment of KIN significantly enhance the R-PC (134.59 ± 5.45 µg g−1 FW and 130.99 ± 7.33 µg g−1 FW) and R-PE (380.44 ± 20.84 µg g−1 FW and 369.90 ± 17.86 µg g−1 FW) content, respectively. TAC also increased under 2-pulse of KIN (630.83 ± 45.12 µg g−1 FW) followed by 3-pulse of combination (592.03 ± 9.73 µg g−1 FW) treatment compared to continuous exposure/1-pulse or control. These findings demonstrate the pulse can enhance the biological responses, offering a more efficient and sustainable approach to seaweed cultivation. To the best of our knowledge, this is the first-ever study on the effect of pulse treatments of bio-stimulants on agarophytic seaweed G. dura. This pulse priming effect would undoubtedly be helpful in enhancing the growth and yield of seaweeds.
