Establishment and characterization of fin cell line from Siniperca chuatsi

The establishment of a cell line is a pivotal advancement in the realm of Siniperca chuatsi breeding particularly in fish diseases research. In this study, we present a novel approach by utilizing caudal fin tissue from S. chuatsi to establish a cell line through tissue cutting. This technique has resulted in the successful creation of the cell line named Scf1, offering a more efficient, clear, and adjustable experimental model that obviates the need for extensive animal usage. The Scf1 cell line derived from S. chuatsi has undergone more than 70 passages in L15 medium supplemented with 20% FBS, exhibiting robust growth at an optimal temperature of 28 °C in the incubator. Upon resuscitation, the Scf1 cell line growth performance is no different from ordinary cells. Through genetic analysis using microsatellite sequences specific to S. chuatsi, it has been conclusively established that the Scf1 cell line originates from this species. Furthermore, experimental infection of the Scf1 cell line with the Micropterus salmoides rhabdovirus (MSRV) resulted in the manifestation of distinct cytopathic effects (CPE), affirming the cell line’s susceptibility to viral infections and its ability to support stable viral replication. Concurrently, transfection of the pEGFP-N2 plasmid into the Scf1 cell line resulted in successful expression, affirming the cell line’s competence as an expression vector. Scf1 stands as the premier cell line isolated from S. chuatsi caudal fins, demonstrating promise as a potent investigative instrument for delving into the molecular intricacies underlying S. chuatsi physiology.
