<p>A 56-day research evaluated the impact of capsanthin intake on the growth, biochemical, and immunological responses of <i>Macrobrachium nipponense</i>. Five nutritional regimens were administered to prawns averaging 1.38 ± 0.06&#xa0;g, each containing varying levels of capsanthin: 0 (control), 0.05, 0.1, 0.15, and 0.2&#xa0;g/kg. Results indicated that growth metrics were notably affected by capsanthin levels (<i>P</i> &lt; 0.05), except for the hepatosomatic index (<i>P</i> &gt; 0.05). Most hemolymph biochemical indices decreased in capsanthin-fed treatments (<i>P</i> &lt; 0.05), while high-density lipoprotein (HDL) and low-density lipoprotein (LDL) levels increased compared to the control group (<i>P</i> &lt; 0.05). Calcium, phosphorus, and cholesterol levels showed no significant differences (<i>P</i> &gt; 0.05). Most hemato-immune parameters, as well as hepatopancreatic antioxidant statuses, demonstrated a significant positive response to capsanthin treatments (<i>P</i> &lt; 0.05). However, glutathione peroxidase (GPx) and alkaline phosphatase (AKP) levels remained unaffected (<i>P</i> &gt; 0.05). A dietary level of 0.2&#xa0;g/kg of capsanthin positively influences digestive enzyme activities, intestinal microflora, total carotenoid content (TCC), and whole-body composition (<i>P</i> &lt; 0.05). The expression of genes involved in growth, immune, and metabolism was influenced by varying capsanthin concentrations (<i>P</i> &lt; 0.05). Prawns receiving capsanthin showed notable enhancements in amino acid and fatty acid profiles, with significant increases in essential amino acids (EAA), monounsaturated fatty acids (MUFA), and polyunsaturated fatty acids (PUFA) compared to the control group (<i>P</i> &lt; 0.05). Overall, findings suggest that adding of 0.2&#xa0;g/kg of capsanthin to the diet of oriental river prawn positively influences their growth, biochemical, immuno-physiological, and metabolic responses.</p>
