Autumnicola aciditolerans sp. nov., a novel member of the genus Autumnicola isolated from tidal flat in Zhoushan

A Gram-stain-negative, aerobic, and rod-shaped bacterium, designated H14T, was isolated from a tidal flat in Zhoushan, Zhejiang Province, China. Colonies of strain H14T were yellow, round, smooth and convex after cultivating on marine agar at 37 ℃ for 72 h. Cells were catalase-positive and oxidase-negative. Growth occurred at 16–40 ℃ (optimum, 37 ℃), pH 5.0–8.5 (optimum, pH 6.5) and with 0.5–10.0% (]/v) NaCl (optimum, 3.0%). Strain H14T showed highest 16S rRNA gene sequence similarity of 97.7% to Autumnicola tepida F363T. It contained menaquinone-6 (MK-6) as the sole respiratory quinone, iso-C15:0 and summed feature 3 (C16:1 ω7c and/or C16:1 ω6c) as the major fatty acids. The main polar lipids were phosphatidylethanolamine (PE), three unidentified aminolipids (AL1-3) and four unidentified lipids (L1-4). The DNA G + C content was 39.6 mol%. The average amino acid identity (AAI), average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between strain H14T and the current genome-sequenced Autumnicola type strains were 78.7–88.4%, 77.1–83.7%, and 17.1–45.8%, respectively. Based on the polyphasic taxonomic analysis, a novel member of the genus Autumnicola, named Autumnicola aciditolerans sp. nov., is proposed. The type strain is H14T (= MCCC 1K08336T = KCTC 82836 T = CGMCC 1.61706 T).
