Morpho-Molecular Characterization and Biocontrol of Fusarium spp. Causing Potato Dry Rot in West Bengal

Potato dry rot is a seed-borne and soil-borne postharvest disease that affects tuber production and is caused by several species of Fusarium. The present study dealt with isolation of five fungal isolates from three potato varieties, ‘Kufri Jyoti’, ‘Kufri Chandramukhi’ and ‘Kufri Pukhraj’, collected from Murshidabad and Nadia districts of West Bengal, India. Three isolates were identified as F. oxysporum and other two isolates as F. sambucinum and F. avenaceum based on conidial morphology and sequencing of rDNA-ITS region. Koch postulates established that each of them alone could incite dry rot symptom in to the susceptible potato tuber and their disease severity in term of tuber tissue penetration varied from 11.89 ± 0.21 mm to 25.10 ± 0.74 mm. The isolates were positive for production of the plant cell-wall-degrading enzymes (pectin lyase, polygalacturonase, cellulase and amylase) and the mycotoxins (deoxynivalenol, fusaric acid, fusarin C, sambutoxin and zearalenones). A T. harzianum (GenBank Accession no. KF826029) isolate was investigated in both in-vitro and in-vivo conditions against all the PDR pathogens. In dual culture, T. harzianum showed percentage of inhibition in the range of 67.6 ± 1.78% (against F. oxysporum P403) to 81.4 ± 2.49% (against F. avenaceum C304). In co-inoculation of tuber, it reduced the disease severity and percentage of effect of application varied from 58.61 ± 0.08% (against F. sambucinum C302) to 70.04 ± 1.12% (against F. oxysporum C205). The antagonist was able to produce chitinase and protease and developed hyphal coiling around pathogen hyphae, and lysed the pathogen hyphae. Thus, the antagonistic isolate could be potentially used against the potato dry rot pathogens.
