Morpho-molecular Characterization of Phyllosticta capitalensis, Causing Leaf Spot of Curry Leaf from West Bengal, India

Curry leaf (Murraya koenigii) is an economically important crop in India due to its culinary and medicinal use. The leaf spot disease of curry leaf caused by Phyllosticta capitalensis was observed in the Medicinal and Aromatic Plants Garden at Kalyani (22°59′21″N latitude and 88°27′18″E longitude), West Bengal, India. The disease severity and incidence were estimated up to 72.98 and 98.24% respectively, during the months of September-October. The disease symptoms were mostly circular to irregular, light brown centre surrounded by dark brown margin. Similar spots were also observed on leaflets and twigs. These spots enlarge, coalesce causing leaf blight and severely infected leaves and twigs had burning appearance. Within infected lesions, there were numerous minute pin head like dark brown to blackish pycnidia. The fungal pathogen was isolated from infected plant parts and its pathogenicity was confirmed through Koch’s postulates. Pycnidia of the pathogen were globose to oval or oblate spheroid measuring 83.11 μm ± 9.31 in diameter. Pycnidiospores (conidia) were unicellular, hyaline, thinned walled nearly oval, broader in middle and tapering at both ends, 8.20 μm ± 1.05 × 5.8 μm ± 0.61 in length and breadth with a single appendage. The ITS region of rDNA was sequenced and phylogenetic analysis with global isolates available in the NCBI GenBank was performed. Based on morpho-molecular analysis, the pathogen was identified as Phyllosticta capitalensis. The pathogen culture on PDA was deposited in National Fungal Culture Collection of India (NFCCI, Accession No. 4906) and the consensus sequence of ITS rDNA region was submitted in NCBI’s GenBank (Accession No. MT832358).
