<p>Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been widely used for clinical diagnoses of human pathogens. However, to date it has been seldom applied against plant pathogens, including viruses. Here, we report a simple identification method for tobamoviruses using MALDI-TOF MS. By employing heat treatment in the presence of formic acid, we obtained effective peptide mass fingerprints derived from coat proteins (CPs) using both highly and simply purified tobamovirus virions. Furthermore, we distinguished the presence of two isolates with amino acid substitutions in CPs. Overall, this method may facilitate the practical diagnosis of plant viruses using MALDI-TOF MS.</p>
