Long-term preservation method for Calonectria ilicicola using barley grains

The viability and virulence of Calonectria ilicicola cultured with barley grains were examined after storage at − 20 °C for up to 19 years. Of 114 isolates, 93.0% remained viable, and their virulence over 19 years was comparable to that at the start of culturing and after 3 years of storage on barley grains. The isolates colonized various parts of the barley grains, but survival was highest in the endosperm. This method is thus highly effective for long-term, stable preservation of C. ilicicola.
