Quantification of pterostilbene in Pterocarpus marsupium heartwood using thin-layer chromatography and high-performance liquid chromatography approaches

Pterocarpus marsupium Roxb. (Vijaysar) is a medicinal plant widely used in Ayurvedic formulations for managing metabolic and inflammatory disorders. The therapeutic potential of its heartwood is linked to various bioactive compounds, yet analytical data for standardization remain limited. In this study, macroscopic, microscopic, thin-layer chromatography (TLC), and high-performance liquid chromatography (HPLC) analyses were carried out to characterize the photochemical profile of the heartwood. TLC and HPLC profiling detected a pterostilbene with its content estimated at 0.3% w/w in the heartwood extract. The developed chromatographic profiles provide supportive data for the quality evaluation and authentication of P. marsupium heartwood. Although the calibration curve demonstrated linearity, its shallow slope highlighted the need for further optimization to improve analytical reliability.
