<p><i>Cassia tora</i> Linn., an annual herb from the Leguminosae family, is widely distributed across Southeast Asia and the Southwest Pacific, often regarded as a significant weed. Traditionally, <i>C. tora</i> has been used in various medicinal preparations for its laxative, antirheumatic, and dermatological properties. Recent studies have also revealed its anti-inflammatory, antifertility, hepatoprotective, and neuroprotective activities, which are owing to the bioactive compounds present in the plant species. The present study aimed to identify and quantify key bioactive compounds: quercetin (QC), gallic acid (GA), epicatechin (EC), and rutin (Rut) in the methanolic extract of <i>C. tora</i> (MECT) leaves and its ethyl acetate fraction using thin-layer chromatography (TLC). Total phenolic content (TPC) and total flavonoid content (TFC) and antioxidant activity assays were employed to select the most active fraction for TLC profiling. Results indicated significant variation in the phytochemical content and antioxidant potential among the fractions, with the ethyl acetate fraction emerging as the most enriched source. Ethyl acetate fraction has more amounts of QC, GA, EC, and Rut compared with the mother methanolic extract. These findings highlight the potential of the ethyl acetate fraction of <i>C. tora</i> as a valuable candidate for the development of pharmaceutical and nutraceutical formulations with potent antioxidant properties.</p>
