The effect of IFNAR2 and TYK2 gene expressions and serum IFN-α levels on disease severity in COVID-19 patients

This study was aimed at determining serum interferon α (IFN-α) levels and TYK2 and IFNAR2 gene expression levels and their relationship to the clinical progression of patients with COVID-19 disease at different clinical severity levels. The study was conducted with 40 patients receiving treatment in intensive care (ICU group) whose quantitative real-time polymerase chain reaction (qRT PCR) test results were positive, 40 patients receiving treatment in the service (service group), 38 polyclinic patients (polyclinic group) and 40 symptomatic patients with negative qRT-PCR tests (PCR (-) group). After total RNA was extracted from peripheral blood samples, the qRT-PCR method was used to determine the expression levels of the TYK2 and IFNAR2 genes. Serum IFN α levels were determined in serum samples by enzyme-linked immunosorbent (ELISA) method. TYK2 expression in participants was significantly decreased in the ICU group when compared with the polyclinic (p = 0.001) and PCR(-) group (p = 0.002) and in the service group when compared with the service and polyclinic (p = 0.031) and PCR(-) group (p = 0.049). IFNAR2 expression was significantly decreased in the ICU group when compared with the service (p = 0.016), polyclinic (p ≤ 0.001) and PCR(-) (p = 0.001) groups. Serum IFN-α levels were highest in the PCR(-) group. When the type-1 interferon response in COVID-19 patients was evaluated based on TYK2 and IFNAR2 expression, it was found to be impaired in the intensive care group; additionally, serum IFN-α levels were low in qRT-PCR-positive patients.
