Hypoglycemic effects of methanolic extract of lemon verbena in streptozotocin-induced diabetic rats

Diabetes is considered one of the most prevalent diseases globally. The American Journal of Managed Care estimates that there will be 1.3 billion diabetics by 2050. Awareness that chemical drugs may have certain adverse effects, the utilization of medicinal plants has garnered significant attention in recent decades. Morocco is renowned for its richness and diversity of medicinal plants, including lemon verbena, which exemplifies its biodiversity and extensive reservoir of bioactive molecules. This work aims to evaluate the hypoglycemic effect of the lemon verbena leaves methanolic extract (LVME) in Wistar rats rendered diabetic by streptozotocin (STZ) injection in the short term by assessing the glucose tolerance test, and the long term (28 days) by evaluating the fasting blood glucose (FBG), clinical signs, biochemical profile, and histopathological alterations. After an intraperitoneal injection of STZ (55 mg/kg), rats with FBG levels ≥ 2 g/L were divided into three groups; diabetic control, LVME group treated with the extract at 225 mg/kg bw, and the group treated with metformin (300 mg/kg bw). Also, the normal group received distilled water for 28 days. FBG and diabetes symptoms were followed along the experimental duration. The animals were euthanized after treatment ended, and their blood and organs were collected for further biochemical and histological examinations. Diabetic rats showed weight loss, polyphagia, polydipsia, and hyperglycemia in the glucose tolerance test and during the 28 days of the experiment. Biochemical analysis revealed also an increase in glycemia, lipids, renal and liver markers, and damaged the liver, kidney, and pancreas tissue. However, LVME treatment decreased both short-term and long-term glycemia, and the treated group gained weight in comparison to the diabetic control group. The treatment impacted positively the levels of biochemical parameters and the damaged tissues. The present study suggests that LVME has good hypoglycemic and hypolipidemic potential to improve the diabetic condition in Wistar rats.
