Effect of diferuloylmethane on neutrophil–lymphocyte ratio, cortisol, and testicular damage in male albino rats co-exposed to thermo-psychological stressors during the hot-humid season

The aim of this study was to evaluate the effects of diferuloylmethane (DFM) on neutrophil–lymphocyte ratio, cortisol, and testicular damage of male albino rats co-exposed to thermo-psychological stressors during the hot-humid season. Then, 25 adult male albino rats were divided into five groups, each consisting of five animals, for this study. Group I (control) was not subjected to any stress, while Group II was exposed to chronic thermal stress (CTS). Group III was exposed to psychological stress (PS) while Group IV was co-exposed to CTS and PS (CTS + PS, only). Group V was pretreated with DFM (200 mg/kg) for 30 days and co-exposed to CTS and PS (CTS + PS + DFM). The rats were euthanized after obtaining blood samples for hematology and sera for analysis of malondialdehyde, cortisol, luteinizing hormone, and testosterone concentrations. Testicular tissues were fixed in Bouin’s solution. A tissue processor, a microtome, and hematoxylin and eosin (H&E) stains were used for tissue preparation to carry out histopathological scoring of the testicles. The results showed that the lowest value of neutrophil-to-lymphocyte ratio (NLR) was obtained in the CTS + PS group, denoting high level of stress, thereby leading to distress and a high rate of negative testicular histopathological changes, which may have been mitigated by pretreatment with DFM. Thus, DFM may have a potential protective effect following exposure to heat and psychological stressors.
