Evaluation of expression level of long non-coding RNA RMRP as diagnostic marker in vitiligo

Vitiligo is a common depigmenting skin disorder characterized by patches of skin losing their pigment. Long non-coding RNAs (lncRNAs) are involved in various biological processes and have been implicated in the pathogenesis of autoimmune and inflammatory diseases.
Aim of the workTo evaluate the serum and tissue levels of the lncRNA Ribonuclease Mitochondrial RNA Processing (RMRP) in vitiligo patients compared to healthy controls and to assess its association with disease severity.
Patients and methodsThe study included 20 vitiligo patients and 20 age- and sex-matched healthy controls. For all participants, a 3 mL venous blood sample was collected for serum isolation and RMRP quantification. Additionally, a 3 mm punch biopsy was obtained from lesional skin in the vitiligo group. Quantitative real-time PCR (qRT-PCR) was used to measure RMRP expression in both serum and tissue samples.
ResultsRMRP expression levels were significantly upregulated in both the serum and tissue of vitiligo patients compared to healthy controls, with p-values of 0.001 and 0.0001, respectively. A strong positive correlation was observed between serum and tissue RMRP levels (r = 0.98, p < 0.0001). In serum, mild and moderate patients had mean expression levels of 20.39 ± 3.92 and 13.61 ± 10.79, respectively. In tissue, the corresponding values were 22.43 ± 4.01 for mild and 15.63 ± 11.49 for moderate cases.
ConclusionThe upregulation of RMRP in vitiligo patients suggests a potential role in disease pathogenesis and supports its promise as a biomarker and a possible therapeutic target.
