Introduction <p>Vitiligo, an autoimmune disorder marked by skin depigmentation, is closely linked to immune dysregulation, including T cell infiltration and pro-inflammatory cytokines. This study explores the potential of baricitinib, a JAK-STAT inhibitor, in promoting repigmentation in vitiligo lesions by modulating immune responses.</p> Methods <p>Using a mouse model of vitiligo induced by hydroquinone, we assessed the effects of baricitinib treatment on lesion repigmentation, CD8 <sup>+</sup> T cell infiltration, T cell populations, and serum TNF-α levels. Immunostaining, flow cytometry, and ELISA were used to analyse these parameters.</p> Results <p>Baricitinib treatment significantly reduced CD8 <sup>+</sup> T cell infiltration in the skin, lowered serum TNF-α levels, and decreased both CD4 <sup>+</sup> and CD8 <sup>+</sup> T cell populations in the blood. Remarkably, these immune modulations correlated with notable repigmentation of the lesions.</p> Conclusion <p>Baricitinib effectively reduces inflammation and T cell infiltration, suggesting it as a promising therapeutic for vitiligo. These findings highlight its potential to modulate immune responses and restore skin pigmentation in vitiligo patients.</p>

Introduction

Vitiligo, an autoimmune disorder marked by skin depigmentation, is closely linked to immune dysregulation, including T cell infiltration and pro-inflammatory cytokines. This study explores the potential of baricitinib, a JAK-STAT inhibitor, in promoting repigmentation in vitiligo lesions by modulating immune responses.

Methods
