<p>Illumination and the application of exogenous plant hormones have been recognized for their roles in modulating the biosynthesis and accumulation of polyphenolic compounds within a variety of plant species. In the present study, the effects of blue light irradiation and methyl jasmonate (MeJA) on the germination and polyphenol content of mung bean sprouts were systematically investigated. Post-extraction, the polyphenolic profile was meticulously quantified using high-performance liquid chromatography (HPLC), while the transcriptional activity of genes pivotal to polyphenol biosynthesis was quantified employing a fluorescence-based quantitative polymerase chain reaction (qPCR) assay. It was observed that under blue light conditions, the polyphenol accumulation in the sprouts peaked at 20&#xa0;μM MeJA, exhibiting 1.78 folds increase relative to sprouts germinated in the dark with an equivalent concentration of MeJA, and 1.20 folds greater than that sprouts under blue light irradiation without MeJA treatment. These results underscore the synergistic effect of MeJA and blue light in enhancing polyphenol accumulation. Concurrently, a marked upregulation in the expression levels of genes associated with polyphenol biosynthesis, specifically <i>ANS</i>, <i>F3H</i>, <i>IF7GT</i>, and <i>3GT</i>, was noted under these experimental conditions. The findings of this investigation offer novel insights into the molecular mechanisms governing polyphenol biosynthesis and suggest a potential strategy for augmenting polyphenol levels in mung bean sprouts through the strategic application of MeJA in conjunction with blue light exposure.</p>
