Fabrication and evaluation of arabinoxylan hydrogels from Hordeum vulgare husk for sustained drug delivery of Itopride HCl

The purpose of the present work was to extract arabinoxylan from Hordeum vulgare husks and exploit it as an excipient in the fabrication of biopolymer-based pH-sensitive hydrogels for sustained delivery of Itopride HCl. Arabinoxylan was efficiently extracted by alkali extraction method and was thoroughly characterized for evaluating its physicochemical properties. It was subsequently employed in the synthesis of hydrogels loaded with Itopride HCl, by means of free-radical polymerization. Various hydrogel formulations were fabricated by varying the concentration of arabinoxylan, and later on their physical properties, including swelling ratio, equilibrium swelling (%), gel–sol content and porosity, were evaluated. The hydrogels demonstrated a pH-dependent response in swelling studies. The FTIR analysis confirmed the formation of polymeric network. SEM, XRD and thermal analysis depicted formation of porous, amorphous and stable hydrogels, respectively. Furthermore, these arabinoxylan-based hydrogels demonstrated sustained drug release for 24 h, which was primarily diffusion controlled. Oral toxicity studies suggested good level of safety of both extracted polymer and AX-based hydrogels. This study concludes that barley husk is an economical source for formulating polymer-based drug delivery systems producing sustained and improved therapeutic effects.
