Herbiconiux dendrobii sp. nov., a Novel Bacterium Isolated from Dendrobium devonianum Sawdust Substrate

In this study, a novel, aerobic, mesophilic bacterial strain capable of siderophore and indole-3-acetic acid (IAA) production, designated YIM B11900T, was isolated and classified. The rod-shaped, Gram-stain-positive bacterium was isolated from rhizosphere sawdust matrix samples of Dendrobium devonianum collected from Longling County, Baoshan City, Yunnan Province, southwest PR China. Strain YIM B11900T exhibited optimal growth at 30 °C (range 10–35 °C), pH 7 (range 5–10), and 1.5% (w/v) NaCl (range 0.5–2.5%, w/v). The 16S rRNA gene sequence similarity analysis revealed that strain YIM B11900T exhibited the highest similarity to strain Herbiconiux oxytropis CPCC 203406T (98.48%). Phylogenetic analysis based on genome data suggested that strain YIM B11900T belonged to the genus Herbiconiux. The average nucleotide identity (ANI) value was 79.7%, the digital DNA-DNA hybridization (dDDH) value was 24.0%, and the average amino acid identity (AAI) value was 77.8% compared to the related strain Herbiconiux oxytropis CPCC 203406T. The major fatty acids were anteiso-C15:0, Summed Feature 8 (C18:1 ɷ7c/C18:1 ɷ6c), and anteiso-C17:0. The polar lipids included diphosphatidylglycerol, phosphatidylglycerol, lipid, and five unidentified glycolipids. The predominant menaquinones were MK-10 and MK-11, and the genomic DNA G + C content was 70%. Considering the polyphasic taxonomic evidence, strain YIM B11900T is proposed as a novel species within the genus Herbiconiux, designated as Herbiconiux dendrobii sp. nov. (type strain YIM B11900T = NBRC 117108T = CGMCC 1.64948T).
