Alicycliphilus soli sp. nov., Isolated from a Plastic Film Greenhouse Soil and the Proposal for the Reclassification of Diaphorobacter limosus as Alicycliphilus limosus comb. nov.

A Gram-stain-negative, aerobic, motile, rod-shaped, pale yellow bacterial strain LB2ST was isolated from a plastic film greenhouse soil in Huaibei, China, and its taxonomic position was investigated using polyphasic approach. Growth occurred at 15–45 ℃ (optimum, 30 °C), at pH 5.0–9.0 (optimum, pH 8.0) and at 0–1.5% (w/v) NaCl (optimum, 0% NaCl). The 16S rRNA gene sequence of strain LB2ST showed high similarity to Oryzisolibacter propanilivorax EPL6T (97.4% similarity), followed by Melaminivora alkalimesophila CY1T (97.1%), Alicycliphilus denitrificans K601T (97.0%), and Paenacidovorax caeni R-24608 T (96.9%). The pairwise average nucleotide identity (ANI), average amino acid identity (AAI) and digital DNA-DNA hybridizations (dDDH) values between strain LB2ST and other Comamonadaceae species were below 85.3%, 85.0% and 29.4%, respectively. Considering the results from phylogeny, phenotypic and genotypic data, strain LB2ST (= CCTCC AB 2023230 T = KCTC 8325 T) represent a novel species of the genus Alicycliphilus, for which the name Alicycliphilus soli sp. nov. is proposed. Furthermore, based on the phylogenomic analysis, it is recommended that Diaphorobacter limosus be reclassified as Alicycliphilus limosus comb. nov.
