The influence of temperature and salinity on motility of Hematodinium perezi dinospores released from the mudflat crabs Helice tientsinensis

Hematodinium is a genus of parasitic dinoflagellates that infect various marine crustaceans, with waterborne transmission of dinospores considered an effective infection pathway. In the coastal areas of the Shandong Peninsula, frequent outbreaks of Hematodinium diseases have significantly affected the sustainable culture of the Chinese swimming crab Portunus trituberculatus. The infectious dinospores released from the wild alternative host, mudflat crab Helice tientsinensis, cause infections in aquaculture ponds, but their activity remains unclear. This study employed cell tracking technology to investigate the effects of temperature and salinity on the motility of Hematodinium perezi microspores and macrospores released from H. tientsinensis. The study recorded the left-handed circular helical trajectories of microspores and macrospores. Low salinity (22) and high temperature (30 °C) significantly inhibited the motility of both microspores and macrospores. In this study, the optimal conditions for maintaining the motility of both types of dinospores were 15 °C and a salinity of 32. This research investigated the temporal changes in the motility of H. perezi dinospores under different environmental conditions, providing valuable insights into the mechanisms of epizootic dynamics in P. trituberculatus aquaculture.
