Purification and characterization of an exopolysaccharide produced by Lactiplantibacillus plantarum YM-4-3 strain isolated from home-made Yunnan Tempeh

In this study, we investigated the enhanced biosynthesis and bioactivity of the exopolysaccharide (EPS) produced by Lactiplantibacillus plantarum YM-4-3, a strain isolated from Yunnan traditional tempeh. Comparative analysis revealed that modified MRS medium (MMRS) significantly increased EPS production from 122.64 ± 1.78 mg/L to 322.47 ± 3.08 mg/L compared to standard MRS culture. Monosaccharide composition analysis demonstrated consistent constituent profiles between culture conditions, though marked differences in relative abundance ratios were observed. By measuring the in vitro and intracellular antioxidant activity of EPS, it was found that the concentration of EPS reached 400 mg/L, the scavenging capacities of EPS produced by MRS Medium and modified MRS Medium reached 11.8 and 12.1% (DPPH), 46.94% and 53.3% (superoxide anion), 18.4% and 22.1% (hydroxyl free radical), respectively. Furthermore, cell-based assays demonstrated that EPS inhibited the growth of HeLa, HCT116, and U2OS cancer cells in a dose-dependent manner after 48 h of treatment. Transcriptomic profiling identified key metabolic pathways associated with EPS biosynthesis, including differential regulation of genes in the pentose phosphate pathway (LPST _ C2661, LPST _ C1728, LPST _ C2936), and galactose metabolism (LPST _ C2851, LPST _ C1737, LPST _ C2855). These findings provide mechanistic insights into the enhanced EPS production under optimized conditions. This study highlights the potential of L. plantarum YM-4-3 EPS as a natural antioxidant and anti-tumor agent, indicating its broad application prospects in the functional food and pharmaceutical sectors.
